Melanin-Concentrating Hormone Receptor Antibodies

Melanin-concentrating hormone (MCH) receptors, primarily MCH1 in humans and MCH2 in some species, are class A G protein-coupled receptors involved in the regulation of energy balance, appetite, mood, and sleep. Pharmacologically, these receptors are activated by the endogenous neuropeptide MCH, while selective antagonists have been developed experimentally as potential treatments for obesity, depression, and anxiety, although no widely approved drugs targeting MCH receptors are currently in clinical use. Upon ligand binding, MCH receptors mainly couple to Gi/o and Gq proteins, leading to inhibition of adenylate cyclase, reduced intracellular cyclic AMP levels, and activation of phospholipase C with subsequent calcium signaling. These signaling pathways influence neuronal excitability, neuroendocrine secretion, and behavioral responses related to feeding and emotional regulation. MCH1 is broadly expressed in the central nervous system, particularly in hypothalamic and limbic regions that control appetite, reward, and mood. Peripheral expression has also been reported in metabolic tissues and the gastrointestinal tract, suggesting additional roles in energy homeostasis. Through their combined pharmacological properties, signaling mechanisms, and expression patterns, MCH receptors represent promising but still largely experimental therapeutic targets in metabolic and neuropsychiatric disorders.
